Differential Effects of Histamine and SRS-A on Airway Function: In Vivo and In Vitro

The effects of histamine and a partially purified preparation of slow-reacting substance of anaphylaxis (SRS-A) on pulmonary mechanics in the guinea pig in vivo and on tissue strip contractile responses in vitro were compared. On the basis of the in vivo data, it was suggested that SRS-A compared to histamine may have selective activity on peripheral airways. This hypothesis was tested by an in vitro evaluation of the contractile responses of tracheal spirals and parenchymal strips to histamine and SRS-A. In this test system, SRS-A when compared to histamine was also found to have selective activity on more peripheral airways, as indicated by parenchymal strip constriction.
