Inorganic Phosphate Measurement in Sea Water
A colorimetric inorganic phosphorus technique heretofore employed with tissue fluids has been adapted for use in sea water. The reagent, containing sulfuric acid, ammonium molybdate, and ascorbic acid, is sensitive to concentrations as low as 0.016 μg-at P/L, and standard
curves have been constructed to cover the following ranges: 0.016 – 2.26 μg-at P/L, using 100 ml samples, and 2.26 – 32.2 μg-at P/L using 10 ml samples.
Freshly prepared ascorbic acid reagent was found to be chemically viable for at least 12 hours. Upon the addition of the proper amount of reagent to an unknown or standard sample, maximum time of color development was found to be between 8 and 12 hours. This color intensity persisted for at least 95 hours. Best results were obtained at room temperature, and all measurements were made in a Beckman Quartz Spectrophotometer, Model DU at wavelength 820 mμ using either 10 cm or 1 cm cuvettes.
