Response of Stylosanthes humilis seeds to 2,4-dichlorophenoxyacetic acid

A study was carried out to investigate the mode of action of 2,4-dichlorophenoxyacetic acid (2,4-D) on germination of seeds of Stylosanthes humilis, an annual tropical forage legume. 2,4-D inhibited S. humilis seeds germination in a dose-dependent manner, with a correspondingly low ethylene production by the seeds. Significant decreases in tricarboxylic acid (TCA) cycle intermediates and amino acids were also observed in seeds incubated with 2,4-D. Notably, the effects of 2,4-D on TCA cycle intermediates and amino acids levels were reverted following transfer of the seeds to 1-aminocyclopropane-1-carboxylic acid (ACC). Furthermore, ethylene production and germination of S. humilis seeds increased following transfer from 2,4-D solution to ACC solution, but not after transfer to deionised water. Together, these combined experiments suggest that 2,4-D inhibited the germination of S. humilis seeds through inhibition of ethylene biosynthesis. These responses were associated with an extensive reprogramming of primary metabolism.
