Disposable Screen-Printed Biosensor Array for Simultaneous Detection of Microcystis and Escherichia Coli Oligonucleotides

In this paper, a disposable screen-printed biosensor array for detecting specific oligonucleotides of Microcystis and Escherichia coli simultaneously is proposed. The array consisted of an Ag/AgCl pseudo-reference electrode, a graphite counter electrode and two gold working electrodes. Thiolated Microcystis and Escherichia coli oligounucleotide probes were covalently immobilized on each surface of gold electrodes respectively, and then target sequences were captured via hybridization with probes, using methylene blue (MB) as the electrochemical indicator. Experiment conditions were optimized to ensure that the detection of DNA hybridization can be successfully achieved in the same buffer. The results indicated that the limits of detection for the biosensor array of Microcystis and Escherichia coli were 2.1 nM and 5.5 nM, respectively. This array showed significant advantages in terms of simplicity, low cost and being efficient to detect different DNA oligonucleotides simultaneously.
