Prostaglandins and leukotrienes are derivatives of unsaturated fatty acids acting as mediators of inflammation. Inhibition of prostaglandin synthesis by nonsteroidal anti-inflammatory drugs accounts for their anti-inflammatory effects in rheumatoid synovium and other tissues, and inhibition of prostaglandin synthesis by glucocorticoids contributes to their anti-inflammatory effects. Endogenous polypeptide factors produced by peripheral blood monocytes, rheumatoid synovium and other tissues stimulate prostaglandin synthesis and may be important in inflammatory processes. Alterations in the fatty acid composition of the diet may lead to changes in production of prostaglandins and leukotrienes, and thereby may be expected to alter inflammatory responses.
