Arachidonic Acid Cascade: The Cyclooxygenase Pathway

Arachidonic acid present in cell membranes is enzymatically converted to prostaglandins of varying biologic activity, depending upon cell type, by the cyclooxygenase pathway. Prostacyclin (PGI2) synthesized by vascular endothelium, and thromboxane A2 (TXA2), synthesized by platelets, have opposing modulatory effects upon hemostasis and vascular tone. The balance between these prostaglandins is affected by plasma factors, availability of arachidonate as substrate, and rates of enzyme inactivation. Aspirin has profound effects upon prostaglandin synthesis by irreversibly acetylating the pivotal enzyme cyclooxygenase, leading to therapeutically useful changes in PGI2-TXA2 interactions.
