Functional dissection of the catalytic mechanism of mammalian RNA polymerase II

High resolution X-ray crystal structures of multisubunit RNA polymerases (RNAP) have contributed to our understanding of transcriptional mechanisms. They also provided a powerful guide for the design of experiments aimed at further characterizing the molecular stages of the transcription reaction. Our laboratory used tandem-affinity peptide purification in native conditions to isolate human RNAP II variants that had site-specific mutations in structural elements located strategically within the enzyme's catalytic center. Both in vitro and in vivo analyses of these mutants revealed novel features of the catalytic mechanisms involving this enzyme.Key words: RNA polymerase II, transcriptional mechanisms, mutational analysis, mRNA synthesis.

La détermination par rayons X à haute résolution des structures cristallographiques des ARN polymérases à sous-unités multiples (RNAP), a éclairé notre compréhension des mécanismes de la transcription. Elles ont aussi fourni un guide important pour la conception d'expériences visant à caractériser davantage les étapes moléculaires de la transcription. Notre laboratoire a utilisé une méthode de purification d'affinité en tandem en conditions natives pour isoler des variants de la RNAP II humaine possédant des mutations ponctuelles dans des éléments structurels localisés stratégiquement au sein du centre catalytique. Les analyses détaillées, in vitro et in vivo, de ces mutants révèlent de nouvelles caractéristiques des mécanismes catalytiques impliquant cette importante enzyme.Mots clés : ARN polymérase II, mécanisme de transcription, analyse de mutation, synthèse de l'ARNm.[Traduit par la Rédaction]
