Fermentation Engineering for Enhanced Paclitaxel Production by Taxus Media Endophytic Fungus MF-5 ( Alternaria sp.)

Taxol is the most widely used antitumor agent that was industrialized in the past decades. It has been globally used for efficient cancer treatment. A 32 fungus was isolated from five-year-old female Taxus yew and male Mantilla yew trees and screened for Taxol production. Only Alternaria alternata MF5 was shown to produce Taxol, as isolated from the bark of female Taxus yew. The fungus was recognized based on the morphological characters, DNA sequencing and Taxol production screening through HPLC. The MZ medium supplemented with 10 mg/L CuSO4, 20 mg/L MeJA and 5 mg/L GA was most effective for high yield of Taxol in the fermenter. The quantity of Taxol by Alternaria alternata MF5 was 5.7 mg/L after 60 h, as quantified by HPLC and found to be 11-fold higher when compared with other findings. Therefore, the isolated fungus can assist as a possible biomass for fungus business, to improve Taxol market production.
