SEPARATION OF POLYPHOSPHATE-ACCUMULATING BACTERIA FROM ACTIVATED SLUDGE SAMPLES USING DENSITY CENTRIFUGATION AND FLOW CYTOMETRY
Two methods to separate polyphosphate-accumulating bacteria from other organisms in activated sludge samples were developed. The first method was based on the differences on buoyant density of microbial cells and resulted in the identification of two groups of PAO with markedly different buoyant densities. The method allowed a 2 to 5 fold concentration of PAO. The second method was based on the fluorescence of DAPI stained cells that contained polyphosphate and the separation of PAO from non PAO using flow cytometry. The resulting sorted samples contained up to 95% PAO. These separation methodologies will be a useful complement to molecular identification methods, since they will allow directing identification efforts to the organisms of interest.
