Free Content Changes in Follicle Cell Epithelium Nuclei at the Onset of Vitellogenesis in the Octopus Eledone Cirrhosa

The process of reproductive maturation has been studied in females of the octopus Eledone cirrhosa. Nuclei of follicle cells surrounding oocytes at a range of maturation stages have been selectively stained for DNA, scanned by microdensitometry, and the DNA content estimated in comparison with sperm nuclei similarly processed. In previtellogenic eggs the DNA content of follicle cells is distributed closely about the diploid cell quantity, twice that of the sperm nucleus. At the onset of vitellogenesis the follicle cells become increasingly polyploid, their nuclear DNA quantities showing peaks of incidence corresponding to a geometrically increasing series up to 4 times the diploid quantity. Although higher and intermediate values are common, the principal mechanism of this increase is interpreted as incomplete cell division. The relationship between these cellular changes and yolk secretion is discussed.
