Colchiploidy of Ulmus pumila and its Possible Use in Hybridization with U. americana

Abstract:

American elm, Ulmus americana, is very susceptible to Dutch elm disease caused by Ceratocystis ulmi, and to phloem necrosis caused by Morsus ulmi. Siberian elm, U. pumila, is highly resistant to both diseases. Previous efforts to hybridize these species in order to obtain trees with growth characteristics of the American elm and with the disease-resistance of Siberian elm have failed, perhaps because of differences in chromosome numbers between the two species. American elm is tetraploid with 4x = 56 chromosomes, and Siberian elm is diploid with 2x = 28 chromosomes. U. pumila seedlings at the cotyledon stage were treated with 0.5 percent colchicine solution, applied on the epicotyl region only. Over half of the seedlings treated were isolated as probable tetraploids in the reproductive tissue.
