An uncommon H3/Ser10 phosphorylation pattern in Cestrum strigilatum (Solanaceae), a species with B chromosomes

Abstract:
Cestrum strigilatum (Solanaceae) is a South American shrub with B chromosomes. Bs show a univalent behavior when a single B is present, have non-Mendelian segregation, and are poor in genes and rich in repetitive DNA. In this study, the histone H3 at serine 10 (H3/Ser10) phosphorylation pattern was investigated during mitosis and meiosis of C. strigilatum collected from the wild and was compared in A and B chromosomes. The results revealed that H3/Ser10 phosphorylation of A chromosomes occurred only in the pericentromeric region in both mitosis and meiosis, whereas in the B univalent, phosphorylation appeared in almost the whole extent of the chromosome, except in the terminal portion of the long arm. In meiosis II, the phosphorylation of A chromosomes was similar to that in the first division of meiosis, but the Bs did not show H3/Ser10 phosphorylation. Our results suggest that phosphorylation at the pericentromeric region may be associated with chromosome motility during cell divisions and with the cohesion of B chromatids in a univalent structure in meiosis I.Le Cestrum strigilatum (Solanacées) est un buisson d'Amérique du Sud qui possède des chromosomes B. Les chromosomes B forment des univalents lorsqu'un seul est présent, affichent une ségrégation non-mendélienne, sont pauvres en gènes et riches en ADN répété. Dans ce travail, la phosphorylation H3/Ser10 est examinée lors de la mitose et la méiose chez le C. strigilatum récolté dans la nature, et la présence de cette modification est comparée chez les chromosomes A et B. Les résultats montrent que la phosphorylation H3/Ser10 chez les chromosomes A était limitée aux régions péricentromériques tant lors de la mitose que de la méiose, tandis que chez l'univalent B, la phosphorylation était présente sur presque tout le chromosome à l'exception de la portion terminale du bras long. Lors de la méiose II, la phosphorylation des chromosomes A était semblable à celle observée lors de la première division de la méiose alors que les chromosomes B ne montraient pas de phosphorylation H3/Ser10. Ces résultats suggèrent que la phosphorylation des régions péricentromériques serait possiblement associée avec la mobilité des chromosomes au cours des divisions cellulaires ainsi qu'avec la cohésion des chromatides B au sein d'un univalent à la méiose I.Document Type: Research article
