Prevention of myosin-induced autoimmune myocarditis in mice by anti-L3T4 monoclonal antibody

Abstract:

This study was aimed at studying the effect of the induction of immune tolerance to swine cardiac myosin from anti-L3T4 monoclonal antibody injection and whether the immune tolerance could protect mice with myosin-induced myocarditis from myocardial injury. Twenty-four Balb/c mice were divided into two groups at random. All of the mice were immunized with swine cardiac myosin on the 1st day, 14th, 28th, 42nd, and 52nd day. Immune tolerance was induced by triplicate injections of 400 <font face="symbol">m</font>g anti-L3T4 McAb on the 0 day (intravenous), 1st day, and 2nd day (intraperitoneal) in McAb-treated group. In the saline-treated group, saline of the same volume as anti-L3T4 monoclonal antibody was used as a control. The sera and hearts biopsies of all mice were collected on the 58th day. The anti-cardiac myosin antibody was examined with ELISA, and pathological changes of heart were observed by light microscope. It was shown that mice immunized with swine cardiac myosin could produce anti-myosin antibody and the anti-cardiac myosin antibody was positive in most of the saline-treated group but negative in the McAb-treated group. Morphologically, myocardial degeneration, necrosis, and infiltration of inflammatory cells were found in the saline-treated group but not in the McAb-treated group. In conclusion, this study indicated that the immune tolerance to cardiac myosin was induced by the anti-L3T4 monoclonal antibody, and accordingly myocardial injury could be prevented by induction of immune tolerance.Key words: anti-L3T4 monoclonal antibody, myosin, immune tolerance, myocarditis.

La présente étude a eu pour but d'examiner l'effet de l'induction d'une tolérance immunitaire par l'anticorps monoclonal (AcMc) anti-L3T4 dans la myocardite induite par la myosine. Vingt-quatre souris Balb/c ont été réparties aléatoirement dans deux groupes. Toutes les souris ont été immunisées avec de la myosine cardiaque de truie les jours 1, 14, 28, 42 et 52. La tolérance immunitaire a été induite par trois injections de 400 <font face="symbol">m</font>g d'anticorps monoclonal anti-L3T4, le jour 0 (intraveineuse), le jour 1 et le jour 2 (intrapéritonéale) chez le groupe traité avec l'AcMc; chez le groupe traité à la solution saline, une solution de même volume que l'anticorps monoclonal ani-L3T4 a été utilisée. Les sérums et les biopsies cardiaques de toutes les souris ont été prélevés le jour 58. L'anticorps anti-myosine cardiaque a été examiné par ELISA et les modifications pathologiques du coeligur ont été observées par microscopie optique. On a constaté que les souris immunisées avec la myosine cardiaque de truie pouvaient produire un anticorps anti-myosine, et que l'anticorps anti-myosine cardiaque était positif chez la plupart des souris du groupe traité à la solution saline, mais négatif chez les souris du groupe traité à l'AcMc. Sur le plan morphologique, une dégénérescence du myocarde, une nécrose et une infiltration des cellules inflammatoires ont été notées chez le groupe traité à la solution saline mais pas chez le groupe traité à l'AcMc. En conclusion, cette étude a indiqué que l'anticorps monoclonal anti-L3T4 pouvait induire une tolérance immunitaire à la myosine cardiaque et que, par conséquent, l'induction d'une tolérance immunitaire pouvait prévenir les lésions myocardiques.Mots clés : anticorps monoclonal anti-L3T4, myosine, tolérance immunitaire, myocardite.[Traduit par la Rédaction]
