Characterization of Streptomyces scabies mutants deficient in melanin biosynthesis

Abstract:

Streptomyces scabies, a causal agent of common scab, produces both melanin and a secondary metabolite called thaxtomin A. To establish a possible relation between melanin and thaxtomin A production in S. scabies, we carried out N-methyl-N′-nitro-N-nitrosoguanidine (NTG) mutagenesis and isolated 11 melanin-negative mutants of S. scabies EF-35. These mutants were characterized for thaxtomin A production, pathogenicity, sporulation, and stress resistance. Nine of these mutants showed a significant reduction in thaxtomin A production when compared with the wild strain. However, only a few mutants exhibited a reduced level of virulence or a loss in their ability to induce common scab symptoms on potato tubers. Other pleiotrophic effects, such as higher sensitivity to heavy metals and incapacity to sporulate under certain stress conditions, were also associated with a deficiency in melanin production.Key words: common scab, potato, secondary metabolism, stress, thaxtomin.

Streptomyces scabies, un des agents responsables de la gale commune, produit de la mélanine aussi bien qu'un métabolite secondaire appelé thaxtomine A. Afin d'établir une éventuelle relation entre la production de mélanine et de thaxtomine A, nous avons effectué une mutagenèse au N-méthyle-N′-nitro-N-nitrosoguanidine (NTG) et avons isolé 11 mutants de S. scabies EF-35 déficients dans la production de mélanine. Ces mutants ont été caractérisés pour la production de thaxtomine A, le pouvoir pathogène, la sporulation et la résistance aux stress. Neuf de ces mutants ont montré une réduction significative de leur production de thaxtomine A comparativement à celle de la souche sauvage. Toutefois, seulement quelques mutants montraient un niveau de virulence réduit ou perdaient leur capacité à induire des symptômes de gale commune sur des tubercules de pomme de terre. D'autres effets pléiotrophiques tels une sensibilité accrue aux métaux lourds et une incapacité à sporuler sous certaines conditions de stress ont été associés à une incapacité à produire de la mélanine.Mots clés : gale commune, pomme de terre, métabolisme secondaire, stress, thaxtomine.
