Polyphasic taxonomy of the basidiomycetous yeast genus Rhodosporidium: R. azoricum sp. nov.

Abstract:

This report presents the description of a new heterothallic Rhodosporidium species, R. azoricum sp. nov. The new species is based on two strains previously identified as Rhodotorula glutinis, which were isolated from soil in São Miguel island, Azores, Portugal. Evidence that the two strains were conspecific and distinct from Rhodotorula glutinis was obtained in DNA fingerprinting experiments using the microsatellite-primed PCR approach (MSP-PCR) and the primers M13 and (GTG)5. In order to determine the phylogenetic position of the new species, the nucleotide sequence of the D1/D2 region of the 26S rDNA was analysed and Rhodosporidium azoricum was found to belong to a cluster including R. fluviale, R. lusitaniae, Sporidiobolus microsporus, and S. ruineniae. The life cycle of R. azoricum was investigated and comparisons integrating physiological, morphological, and molecular data were made with related species.Key words: Rhodosporidium azoricum sp. nov., microsatellite-primed PCR, Rhodotorula glutinis, basidiomycetous yeasts.

Nous présentons dans ce rapport la description d'une nouvelle espèce hétérothallique de Rhodosporidium, R. azoricum sp. nov. L'existence de cette nouvelle espèce se base sur deux souches identifiées auparavant comme Rhodotorula glutinis, qui ont été isolées à partir du sol de l'île de São Miguel, Açores, Portugal. La preuve que les deux souches étaient conspécifiques et distinctes de Rhodotorula glutinis fut obtenue par des prises d'empreintes génétiques en utilisant l'approche du PCR amorcé par microsatellites (MSP-PCR) et les amorces M13 et (GTG)5. Afin de déterminer l'emplacement phylogénétique de la nouvelle espèce, la séquence de nucléotides de la région D1/D2 de l'ADNr 26S a été analysée. Rhodosporidium azoricum a démontré une appartenance à un groupe comprenant également R. fluviale, R. lusitaniae, Sporidiobolus microsporus et S. ruineniae. Le cycle vital de R. azoricum a été examiné et des comparaisons incorporant des données physiologiques, morphologiques et moléculaires ont été faites par rapport à des espèces apparentées.Mots clés : Rhodosporidium azoricum sp. nov., PCR amorcé par microsatellites, Rhodotorula glutinis, levures basidiomycètes.[Traduit par la Rédaction]
