Objective: To study the effect of gastrodin on acetylcholine release in rat brain. Methods: A liquid chromatography-tandem mass spectrometry method was developed and validated for the determination of acetylcholine in rat brain dialysates. The dialysates were prepared by microdialysis before and after subcutaneous administration of gastrodin at different dosages. Results: The results indicated that gastrodin significantly increased acetylcholine release in rat medial prefrontal cortex, nucleus accumbens and hippocampus in a dose-related manner. Conclusion: The mechanism of gastrodins action in the brain may be related to the regulation of cholinergic neurotransmitter release.
