Analysis of saponins in black red ginseng by HPLC-MS/MS

Objective: To analyze and study the contents of saponins in the nanometer black red ginseng and the common black red ginseng by HPLC-MS/MS and lay a foundation for further development of the product of fermented black red ginseng. Methods: Saponins in the nanometer black red ginseng and the common black red ginseng were extracted by methanol with the Soxhlet apparatus,then were concentrated and filtered by 0.22-μm membrane.An Agilent XDB C18(3.0 mm×100 mm,1.8 μm) column was used for liquid chromatography,with 0.05% aqueous formic acid and methanol as the mobile phase for gradient elution (0-0.5 min,20%B;0.5-5 min,20%B→50%B;5-45 min,50%B→80%B;45-50 min,80%B→100%B;50.1-55 min,20%B),with a flow rate of 0.4 mL·min-1,and a column temperature of 55 .The mass spectrometry detection was performed with electrospray ionization and negative ion in multiple reaction monitoring (MRM) mode to determine the contents of saponins in black red ginseng. Results: Fifty-seven ginsenosides in the nanometer black red ginseng and 30 ginsenosides in the common black red ginseng was determined by LC-MS/MS,respectively.Among them,2 kinds of ginseng aglycone and 2 acetylated ginsenosides were determined in the nanometer black red ginseng,while none was detected in the common black red ginseng. Conclusion: There are more rare ginsenosides in the black red ginseng,and there are more types and higher contents of saponins in the nanometer black red ginseng than those in the common black red ginseng.
