Study on HPLC specific chromatogram of processed pieces of Sophorae Flos

Objective: To explore the effect on constituents of Sophorae Flos by processing,according to the specific chromatogram of Sophorae Flos,Sophorae Flos Tostus and Flos Sophorae Carbonisatum by HPLC. Methods: Resin was used in the preparation of samples,and the residue of the three pieces of Sophorae Flos was detected by HPLC.The column was a reverse-phase column(Kromasil C18,5 μm,4.6 mm×250 mm).Separation was performed by linear gradient elution using organic phase and water phase.The organic phase for 20% and 50% ethanol eluates was a mixture of methanol,acetonitrile and (5: 1: 0.1) containing glacial acetic acid(0.4%),and for 95% ethanol eluates was methanol containing acetic acid(0.4%).The water phase was water containing glacial acetic acid(0.4%).The flow rate was 0.8 mL·min-1.The column temperature was set at 320 nm.Data analysis was performed by a professional software named Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2004 A). Results: The common peaks of specific chromatogram in the three ethanol eluates of Sophorae Flos,Sophorae Flos Tostus and Sophorae Flos Carbonisatum were 12,12,18;10,10,8;18,18,12;separately.The similarities was over 0.9 between Sophorae Flos,Sophorae Flos Tostus and was below 0.6 between Sophorae Flos and Sophorae Flos Carbonisatum. Conclusion: The series and contents of constituents in the Sophorae Flos Tostus were same as Sophorae Flos.After carbonized,the content and composition of the constituents of Sophorae Flos changed significantly.
