Optimization of separation condition for TCM Pueraria thomsonii's chromatographic fingerprint from different gradient profile
Objective: In order to obtain TCM's chromatographic fingerprint separation with good sensitivity,feasibility and reproducibility.The optimization strategy of chromatographic separation condition was found. Method: TCM Pueraria thomsonii was taken for example in
this paper.The grid search algorithm was employed to optimize the separation condition of different gradient profiles,such as sample linear gradient,multi-segments linear gradient and mixed gradient.The advantages and disadvantages were also compared. Results: The results showed that 14 constituents
of herbal medicine Pueraria thomsonii extracts were separated well under the mixed gradient modes.Moreover,the analysis time was short and the chromatogram separation was satisfying but numbers of optimization parameters was much more and optimization process was more
difficult.While the number of optimization parameters is fewer but the chromatogram separation is not satisfying sometimes under the linear gradient elution.And the situation of the multi-segments linear gradient profile is between the above the two modes. Conclusion: In sum,the single linear
gradient profile is usually preferred to separate complicated compounds in herbal medicine.The complex gradient modes could be applied when it couldn’t meet the requirements of the separation.
