HPLC determination of rutin and quercetin in buckwheat husks

To develop an HPLC method for determination of rutin and quercetin in buckwheat husks.Methods:A Zorbax SB-C18column (4.6 mm×150 mm5 μm) was used with a mobile phase of methanol-0.4% phosphoric acid solution (50:50),the flow rate was 1.0 mL·min-1,the UV detector was set at 254 nm.Results:A linear relationship of rutin was good at the range of 0.024-0.963 mg·mL-1 (r=0.9996) and quercetin was good at the range of 0.0132-0.528 mg·mL-1 (r=0.9990).The average recovery of rutin was 100.1% (RSD=1.15%) and of quercetin was 99.9%(RSD=1.04%).Conclusion:The method is simple,accurate and good reproducibility.It is suitable for the simultaneous determination of rutin and quercetin in buckwheat husks.
