RP-HPLC Determination of Effects of Isoliquiritigenin on Brain Energy Metabolism in Repeated Cerebral Ischemia-reperfusion Mice

To establish an RP-HPLC method for determining the contents of adenine nucleotides in tissue samples and to study the effects of isoliquiritigeninISLon brain energy metabolism by this RP-HPLC method in repeated cerebral ischemia-reperfusion mice. MethodsThe repeated cerebral ischimia-reperfusion models in mice were made by repeated occlusion of bilateral common carotid arteries and reperfusion. The levels of ATPenergy chargeECand total adenine nucleotidesTANof the brain energy state were analyzed by RP-HPLC. The Hypersil C18(4.6 mm×250 mm5 μmcolumn was usedthe mobile phase was 100 mmol·L-1 KH2PO4- K2HPO4buffer solutionpH 6.0)the flow rate was 0.7 mL·min-1the column temperature was 25 the detection wavelength was 254 nm.ResultsThe linear range was 1.7-212.0 μg·mL-1(r=0.9999for ATP1.6208.0 μg·mL-1(r=0.9999for ADP1.3164.0 μg·mL-1(r=0.9999for AMP. The average recoveriesn=6were 101.3%(RSD=1.0%for ATP99.0%(RSD=1.4%for ADP97.6%(RSD=1.6%for AMP. ISL(102040 mg·kg-1·d-1increased the levels of ATPEC and TAN in a dose-dependent manner in repeated cerebral ischemia-reperfusion mice brain.ConclusionThis analytical method is simpleaccurate and reliable. ISL can significantly improve the brain energy metabolism in repeated cerebral ischemia-reperfusion mice.
