HPLC Determination of Gallic Acid and Chlorogenic Acid in Polygonum bistorta L.

To develop an HPLC method for the determination of gallic acid and chlorogenic acid in Polygonum bistorta L.Method:A Diamonsil C18 column(5 μm,250 mm 4.6 mm)was used with a mobile phase of methanol-0.4% acetic acid(25:75)and flow rate of 0.8 mL·min-1.The detection was set at 290 nm and the column temperature at room temperature.Results:The recoveries of gallic acid and chlorogenic acid were 95.7%-104% and 96.0%-100%,respectively.The linear ranges were 5.3-53 μg·mL-1 with a regression coefficient of 0.9996 for gallic acid and 25-150 μg·mL-1 with a regression coefficient of 0.9998 for chlorogenic acid.Conclusion:The method described in this paper is simple and accurate with good reproducibility and can be adopted for the quality control of Polygonum bistorta L.
