Determination of Zolmitriptan in Human Plasma byRP-HPLC with Liquid-liquid Extraction
To establish a RP-HPLC method for determination of zolmitriptanZOLin human plasma.Methods:ZOL and internal standard(tinidazole,TDZ)were extracted from plasma with dichloromethane.The compounds were separated on a Hypersil C18 column(200 mm×5.0 mm5 μm).The mobile phase was a mixture of acetonitrile-0.05 mol·mL-1NaH2PO4 in a ratio of 15:85(adjusted pH to 5.0 by H3PO4)the flow rate was 1.0 mL min-1.The column temperature was room temperature.The detection wavelength was UV 223 nm.Results:A good linearity was obtained from 1.92 to 66.14 ng·mL-1in plasma.The lowest concentration detected was 1.92 ng·mL-1.The recovery of method were from 90.62%to 105.7%and recovery of extraction were from 71.70%to 114.75%.Conclusion:The mrthod is sensitivea accurate and reproducible with good recovery.
