RP-HPLC Determination of Midazolam in Plasma

A reversedphase high performance liquid chromatographic method for the determination of midazolam in plasma is described in this paper.Diazepam was used as the internal standard.Midazolam was extracted from alkalized plasma with chloroform and evaporated to dryness.HPLC analysis was carried out on μ Bondpak C18 column with methanol-water-acetonitrile as the mobile phase and UV detection at 220nm.The lower limit of detection of midazolam in plasma is 3ng/ml,calibration curves is linear over the concentration range of 5500ng/ml.The assay procedure has been applied to monitoring plasma levels in intravenous infusion in 7 volunteers.The results show that the method is suitable for clinical pharmacokinetic research.
