Cytokinin stimulation of seed germination in Rorippa subumbellata Rollins

Abstract:

Seeds of Rorippa subumbellata Rollins, a rare species from the shores of Lake Tahoe (Nevada and California, US), did not germinate when stratified and exposed to two temperature regimes routinely used for seed germination. In order to manage this species ex situ, in vitro methods of germination were examined. When seeds were surface sterilized and incubated on nutrient medium in vitro, little or no germination occurred in the presence of half-strength or full-strength MS medium with 1.5% or 3% sucrose, respectively. However, 88% of seeds on MS medium containing 0.5 mg/L BAP with and without 0.05 mg/L NAA germinated. Although only a few seeds were available for these experiments, the application of the cytokinin, BAP, appears to overcome dormancy in this species and allow for germination ex situ.
