Abstract:

<bold>Evaluation of: Petersen RC, Thomas RG, Grundman M et al.: Vitamin E and donepezil for the treatment of mild cognitive impairment. N. Engl. J. Med. 352, 2379–2388 (2005).</bold> A 3-year randomized controlled study comparing donepezil with tocopherol and placebo in 769 subjects with amnestic mild cognitive impairment showed an overall 16% yearly rate of progression to dementia. This was more common in the subjects carrying the apoE4 allele. There was no sustained difference between the treatment groups over the 3 years of treatment. This study design can be used to test new agents that may modify the progression of Alzheimer’s disease.
