Water-soluble vital stains administered to adult Manduca sexta (L.) by feeding or by injection into the haemoroel were effective in marking spermatophores. However, injection was the more efficient technique and did not damage the moths. Of more than 60 stains that were screened, rhodamine B, methylene blue chloride, pararosaniline acetate, and pararosaniline hydrochloride were the only dyes which stained the spermatophore. Well-stained spermatophores were transferred by males that had been treated with rhodamine B 72 hours before the males were allowed an opportunity to mate.
