The effectiveness of sonication for solubilizing cholinesterase (ChE) was determined with homogenates of thoraces from the house fly, Musca domestica L. The level of increase in ChE activity in the supernatant fraction and the stoichiometric decrease in the pellet fraction after low-speed centrifugation were related to the duration of sonication. Based on ultra-centrifugation and gel filtration experiments, however, the increased activity in the supernatant following sonication and low-speed centrifugation did not represent solubilization of ChE but rather fragmentation of membranes into smaller particles which may appear soluble under some experimental conditions.
