Genotyping of polymorphic marker (MSP3α and MSP3β) genes of Plasmodium vivax field isolates from malaria endemic of Thailand

Abstract:

Summary Two polymorphic marker genes, merozoite surface protein 3α () and merozoite surface protein 3β (), from 100 field isolates, were investigated using polymerase chain reaction and restriction fragment length polymorphism (PCR/RFLP). Genotyping of and revealed marked polymorphisms in length and sequence. Three major types of (Type A, B and C) and two major types of (Type A and B) were detected based on the length of PCR products. Fourteen alleles of both genes with difference frequencies were distinguished by restriction fragment length polymorphism, and these results strongly support that isolates in Thailand are markedly diverse. and are reliable polymorphic markers for population genetic analysis of , and PCR/RFLP provides a powerful method for genotyping and identification of mixed parasite infections without requirement of gene sequencing.

Deux gènes marqueurs polymorphes de la protéine de surface 3α (PvMSP3α) et la protéine de surface 3β (PvMSP3β) du mérozoïte, provenant de 100 isolats de Plasmodium vivax de terrain ont étéétudiés par PCR-RFLP. Le génotypage de PvMSP3α et PvMSP3β a révélé d’importants polymorphismes de taille et de séquence. Trois types majeurs de PvMSP3α (Type A, B et C) et deux types majeurs de PvMSP3β (Type A et B) ont été détectés sur base de la taille des produits de PCR. 14 allèles des deux gènes avec des différences de fréquence ont été distingués par le polymorphisme de la taille des fragments de restriction. Ces résultats soutiennent fortement que les isolats de P. vivax de Thaïlande sont significativement différents. PvMSP3α et PvMSP3β sont de fiables marqueurs polymorphes pour l’analyse génétique des populations de P. vivax et la PCR/RFLP fournit une méthode puissante pour le génotypage et l’identification des infections parasitaires mixtes sans la nécessité de séquençage des gènes.
